The uptake of P-32 into phosphorus compounds of retinal pigment epithelium and retina was investigated in vitro. In both tissues, the rate of P-32 uptake into acid-soluble organic phosphorus compounds ivas considerably greater than into phospholipid or nucleic acid. The corresponding uptake into each of the fractions was the same for both tissues. The rate of P-32 uptake into acid-soluble organic phosphorus of retina did not increase ivith incubation time between 1 and 4 hours, but there was a significant increase in rate of uptake by pigment epithelium to a plateau at 3 hours. Addition of 1 mg. diaminodiphenoxyheptane (DDH) to the tissue preparations before incubation caused a profound decrease in P-32 uptake into acidsoluble organic phosphorus at all incubation times, but no significant decrease in P-32 uptake into the corresponding fraction of retina. A specific inhibitory effect on pigment epithelium is indicated at the level of either enzyme or cell membrane activity.
