A method has been devised for counting and size determination of all the fibers in an optic nerve cross-section. The method employs flying spot scanning of a series of 600 to 700 35 mm. negatives which in composite represent the complete cross-section of the optic nerve. The results of absolute counts on two human and two rhesus monkey optic nerves are reported here. These results show that only the highest figures reported by others in the past on the basis of sampling are the correct figures. The number of fibers in the rhesus optic nerve is in the range of 1.5 to 1.8 million. The corresponding figure for the human lies between 1.1 and 1.3 million. Both the modest variability between individuals, and the larger number of fibers in the monkey than in the human appear to be real. Size information will be presented in a subsequent publication.
